Description
T4 Phage β-glucosyltransferase is a DNA-modifying enzyme encoded by bacteriophage T4 that specifically transfers the glucose moiety of uridine diphosphoglucose (UDP-Glc) to the 5-hydroxymethylcytosine (5-hmC) residues in double-stranded DNA, resulting in the formation of beta-glucosyl-5-hydroxymethylcytosine.

Unit Definition
One unit is the amount of T4 Beta-glucosyltransferase required to protect 0.5 ?g of fully 5-hydroxymethylated 1 kb PCR fragment from cleavage by EcoR 1.
Components
T4 Beta-glucosyltransferase:
10,000 units/ml in 10 mM Tris (pH 7.5 at 25°C), 500 mM NaCl, 0.1 mM EDTA, 1 mM DTT and 50% (v/v) glycerol.
10X Reaction Buffer
: 330 mM Tris-acetate (pH 7.5), 660 mM potassium acetate, 100 mM magnesium acetate, and 5 mM DTT.
