Description:
IL-6 Responsive Luciferase Reporter HEK293 Cell Line is derived human embryonic kidney, and stably express firefly luciferase reporter gene under the control of IL-6 response element. This cell line is an ideal cellular model for monitoring the activation of MAPKs, PI3Ks, STATs response Pathway triggered by stimuli treatment, enforced gene expression and gene knockdown.
Principle:
Interleukin-6 (IL-6) is a multifunctional cytokine which plays an important role in a wide range of
BIOLOG
ic processes in different types of cell including cancer cells. IL6 can stimulate multiple pathways involving MAPKs, PI3Ks, STATs, and other signaling proteins, and subsequently activate its downstream targets in the regulation of cell proliferation, survival, and metabolism. Even though IL6 was originally characterized as a regulator of immune and inflammatory responses, elevated expression and activities of IL-6 has been detected in multiple cancers, such as breast, lung and lymphoma.? IL-6 was associated with tumor progression through increasing antiapoptotic activity, stimulation of angiogenesis, and drug resistance. The increased serum IL-6 concentrations in patients are linked to the advanced tumor stages of various cancers and short survival rate. Developing drugs to block pathological IL-6 overproduction has become a potential therapeutic strategy for cancers.

Principle behind TF luciferase reporter.
? TF luciferase reporter stable cell line utilizes artificial promoter constructs to drive luciferase expression.? The promoter region can consists of multiple repeats of a cis-element TF binding site, a DNA fragment from the promoter region of a known TF downstream gene, or a DNA fragment containing putative/known TF binding sites.? There are several ways that a TF can be activated, such as through extracellular stimuli or through intracellular signaling pathways.? Once activated, the TF translocates to the nucleus and often interacts with relevant co-factors to drive gene expression.? Once luciferase is expressed, it can generate light in an enzymatic assay and the amount of light measured is positively correlated with the level of TF activation.

Analysis of SL-0048 IL-6 reporter activity in response to Differnt cytokine treatment.?
The cells were seeded on a 96-well plate in 10% FBS DMED medium. The cells then immediately were treated with or without 10ng/ml IL-6, IL-15, IL-8, IFNr, IL-6 TNFα in DMEM and 10% FBS for 24 hours. IL-6? induced over 40-fold increase in luciferase activity compared to untreated cells.
