Mesenchymal Cell Preparation and Use
Human Bone Marrow-Derived MSCs are generated by culture-expansion of BM MNCs on tissue culture-treated plasticware in
Reachbio
’s MSC Complete Medium (catalog #2121). Depending upon the growth characteristics of the particular batch, they are harvested at passage number 1, 2 or 3 (ask for Certificate of Analysis for batch-specific passage number). The cells are vialed and cryopreserved in a serum-free freezing medium at 0.5 million cells/mL. Upon thawing, the MSCs can be cultured in MSC Complete Medium and passaged another 6-8 times.

Bone Marrow Sample Collection and Processing
Human bone marrow (BM) MNCs are obtained by density centrifugation of BM cells.?? BM cells are drawn from the posterior iliac crest of normal human donors using a syringe containing anticoagulant.? BM cells are processed within 24-30 hours after collection. Donors have been screened to be negative for HIV, HBV, and HCV.

Pre-Qualified by Flow Cytometry
Each batch of BM-derived MSCs is pre-qualified by flow cytometry to ensure the cells express the proper cell surface phenotype characteristic of MSCs. ?? The cells are verified to express CD29, CD73, CD90 and CD105 but lack expression of the hematopoietic surface

BM-derived MSCs are cryopreserved in CryoStor? CS5, a serum-free freezing medium. Cells are stable for long-term storage at -152

C or in the gaseous phase of liquid nitrogen. Storage at -80

C is not recommended.? Once thawed, the cells must be used immediately and have a finite life span in culture.
