Basic ELISpot for enumeration of IgM secreting B cell
Mouse IgM ELISpot
BASIC
enables enumeration of B cells secreting total mouse IgM as well as antigen-specific mouse IgM. It contains matched quantities of monoclonal capture and detection antibodies, Streptavidin-HRP and polyclonal activators.
Antigen-specific B cells can be enumerated in two alternative ways; by coating with antigen or by using biotinylated antigen in the detection step. For memory B cells, we recommend pre-stimulation with R848 and recombinant mouse IL-2 (both included).

Analyte Information

IgM
Monomeric IgM (Immunoglobulin M) is found on the surface of B cells whereas the IgM molecules secreted from plasma B cells are pentamers, i.e. five IgM molecules joined together by the J chain. IgM constitutes approximately 10% of the total Ig concentration in serum. IgM is the first antibody isotype induced in a typical immune response and hence is the isotype with the lowest affinity for antigens.
