This
ELISA
based assay measures the loss of biotinylated poly (ADP-ribose) from immobilized histones in a 96-well format due to PARG activity. The addition of Strep-HRP (biotin-binding protein) and a chemiluminescent HRP substrate yields relative light units (RLU) that correlates with PARG activity.
