The polylinkers found in M13 and pUC cloning vectors contain a single AccI site.AccI requires at least 13 base pairs beyond the end of its recognition sequence to cleave efficiently.This enzyme has shown to have lower activity on some supercoiled plasmids, with more than 1 unit required to digest 1 μg plasmid DNA.
